TEM # 945-4 S. pseudopedicellatum
Excised samples were fixed in 2% glutaraldehyde, 1% osmium tetroxide,
in 0.1M sodium cacodylate buffer, pH 6.8, for 15 min, then 2 changes of 2%
glutaraldehyde, 2% osmium tetroxide in buffer for 15 minutes. They were
rinsed in 3 changes of water, stained en bloc in 0.5% uranyl acetate in
the dark for 60 min, dehydrated in an ethanol series, infiltrated in an LR
White resin series overnight, flat embedded in resin, sectioned, stained with
Reynold’s lead citrate, and imaged with a JEOL 100CX TEM.
